BackGround:A proliferation-inducing ligand (APRIL), also designatedTNFSF13, is a Type II membrane protein that sharescharacteristics with other members of the tumor necrosisfactor (TNF) cytokine family. APRIL is expressed in highlevels in transformed cell lines and in human colon, thyroid,and lymphoid tumor tissues. APRIL is critically involvedin the regulation of infections, inflammation, autoimmunediseases, and tissue homeostasis. APRIL is implicatedin the regulation of tumor cell growth. TheC-terminal extracellular domain has b jelly roll topographyand is important in ligand trimerization. The bindingof the ligand to its respective receptor induces oligomerization,initiating downstream signaling events. Intrinsic tooligomerization is the formation of the receptor bindingsite at the junction between neighboring subunits, creatinga multivalent ligand.
